Welcome to the ADVIA® 2120i Hematology System Hardware and Software Overview.  The ADVIA® 2120i is a fully automated diagnostic hematology instrument. The analyzer provides results for five selectivities: Complete Blood Count (CBC) CBC plus white cell differential counts (CBC/diff) Reticulocyte counts (retic) CBC/diff plus retic (CBC/diff/retic) CBC/retic Samples are aspirated in one of three modes and are processed using Unifluidics technology. There are five channels for measurement, Hemoglobin, RBC/Platelet, Retic, Baso and Perox. Each Channel has its own unique chemical reaction. The analyzer has a throughput of 120 samples per hour for CBC/diff and 74 samples per hour with a reticulocyte count. The instrument provides results for four selectivities; Complete Blood Count, a CBC plus white cell differential count, Reticulocyte counts, CBC diff plus retic and a complete blood count with retic. Three modes can be used for sample aspiration, Manual Open Tube Sampler (MOTS), Manual Closed Tube Sampler (MCTS) and the Autosampler. The samples are processed using Unifluidics technology then counted and measured using the 3 optical assemblies which consist of the Hemoglobin Colorimeter, Perox Optical Assembly and the RBC Optical Assembly. The system utilizes 5 measurement channels ,Hemoglobin, RBC, Platelet, Retic, Baso,  and Perox.  Each channel has its own unique chemical reaction. Other key components include the Sample and Sheath syringes, Touchpad,  Manual Barcode Reader and the Waste removal system. List the three modes of aspiration The analyzer can have up to three modes of aspiration: Automated closed tube mode: Utilizes the autosampler, which automatically transports, mixes, identifies and aspirates samples in closed tubes. Manual close- tube mode (MCTS) -- To aspirate a sample using the manual closed tube sampler (MCTS), the capped vial is inserted upside down into the centering collar.  The sample tube is then pressed down into the centering collar. Manual open-tube mode (MOTS): To aspirate a sample using the manual open tube sampler (MOTS), the sample probe is first immersed into the specimen.  The sampling process is initiated when the aspirate plate located directly behind the sample probe is pressed.  The  manual modes allow for the introduction of stat specimens and specimens with small sample volumes.  All three modes require that only a minimal sample volume of 175µl be aspirated. Locate all Reagents All reagents are located on the instrument with the exception of SHEATH/RINSE. Sheath rinse is typically located on the floor below the analyzer. Some reagents are packaged together to simplify reagent handling. They are referred to as TIMEPAC™ reagents and are replaced as a unit. Navigate the system software The ADVIA®  2120i system software consists of tabs, menus, and Online Help Menus: Each of the Menu buttons on the top of the screen displays a menu of system functions. The menu displays when the pointer is moved over the button. When selecting an item from the Menu, a corresponding window opens and other functions display as tabs.   Tabs: Correspond to items on the active Menu button. You can switch to any of the functions by selecting the corresponding tab. When an option from a different menu is selected, the tabs change to reflect those of the active menu. Online Help: A resource tool for operators of the ADVIA 2120i Hematology System Is an electronic version of the Operator’s Guide Provides easy access to instructions, descriptions, and theory of system operation Locate Online Help The online Operator's Guide can be accessed using the Info menu button, located at the top right area of the screen. Each screen in the system software contains a Help button, which also provides a link to the Operator’s Guide. When the link is selected, information for that particular screen displays. The Operator's Guide should not be minimized and should be closed by selecting the "X" in the box at the top right corner of the screen. Base ImageHotspotsText BlocksImage File Autosampler Manual Open Tube Sampler (MOTS)   Manual Closed Tube Sampler (MCTS)  Unified Fluids Circuit (UFC)  The UFC is made of eight acrylic plates. Within the fused plates are the pathways for fluids and air flow, valves and four reaction chambers. The Perox reaction chamber and the sample shear valve (SSV) are mounted on the front surface of the UFC block. The reagent pump assembly is mounted to the bottom of the UFC.     Perox optical assembly  RBC optical assembly  HGB colorimeter  Sample and sheath syringes  The perox sample and sheath pumps are located on the left side of the analyzer. The RBC/baso/retic sample and sheath pumps are located on the right side of the analyzer.   Touchpad  The touchpad is located on the lower right of the analyzer.  It is used to operate the analyzer and the buttons toggle. "On" provides power to the analyzer "Standby" sets the analyzer to a lower power state "Start/Stop Sampler" starts the autosampler "Eject Rack" Moves all racks in the autosampler into the output queue "Rack in Sampler" lights up when a sample rack is in the autosampler "Off" turns off the power to the analyzer    Manual Barcode Reader  The manual barcode reader is used to enter information from the labels on sample tubes, reagent containers, controls, and calibrators. As each label is correctly read, the LED on the wand will blink.   Waste Removal System  Analyzer waste is stored in an 11-liter waste container. The waste container can be a stand-alone container which is emptied manually or an automatic waste container.    SHEATH/RINSE SHEATH/RINSE is located near the instrument.  Sheath is a fluid that envelops the sample stream as it passes through the optics ensuring cell-by-cell analysis. All of the reagents are located on the instrument with the exception of SHEATH/RINSE, which is located below the analyzer.  Some reagents are banded together and are referred to as a TIMEPAC™.  To simplify reagent handling these TIMEPAC reagents are replaced as a unit. The remainder of the reagents are replaced individually. The ADVIA 2120i Operator's Guide is a part of the system software.  It provides easy access to instructions, descriptions, and theory of operation.  The Operator's Guide can be accessed from the Info menu button. Tab TitleTextMain Screen There are several ways to access information using the Operator's Guide.  Information can be accessed using the Contents, Index and Search function tabs located at the top of the screen. The Operator's Guide should not be minimized and should be closed by selecting the "X" in the box at the top right corner of the screen   Contents The Contents tab functions like chapters in a book, which can be expanded to view the subchapters. The Contents function can be useful when there is general knowledge of where the information can found.   Index The Index tab allows a keyword to be entered.  In this example "calibr" was the keyword entered.  When the word calibration is selected in the information box, the main topics for the keyword "calibration" appear in a drop down box.   Search When you enter a keyword using the Search function tab, the information box contains every piece of information in which the keyword appeared.   Glossary The Glossary provides the definition for key terms used on the ADVIA® Hematology system.   Hypertext The Operator's Guide contains hypertext links, which when selected will provide the operator with additional information related to the content on the screen.   Help The Operator's Guide can also be accessed by selecting the Help button, which appears on each screen in the system software. The Help button provides a link to the Operator's Guide and additional information about the current screen.   When complete, select the X in the upper-right corner to close the window and continue.   The analyzer has three optical assemblies which consist of: Perox optical assembly RBC optical assembly Hemoglobin colorimeter assembly Optical Assemblies Learn about the Optical Assemblies Tab TitleTextPerox Optical Assembly The perox optical assembly directs light from a tungsten-halogen lamp to the perox flow cell. The use of a sheath stream in the flowcell, allows a cell-by-cell measurement of light scatter and absorption. The perox optical assembly measures the size and staining activity of the WBC's and provides a differential count as well as a secondary white count.  RBC Optical Assembly The RBC optical assembly utilizes a laser diode light source. The use of a sheath stream in the RBC flowcell allows a cell-by-cell measurement of low and high angle light scattering and absorption.  This optical assembly is shared by the RBC/Plt, retic and baso lobularity channels.  HGB colorimeter The hemoglobin colorimeter takes voltage readings which correspond to the amount of transmitted light that passes through the reaction chamber The hemoglobin concentration is then calculated using baseline and sample readings taken at specific intervals during the analysis period. Tab TitleTextAutosampler The autosampler automatically transports, mixes, identifies and aspirates samples in closed tubes. The autosampler is a rack style sampler which can accommodate multiple size tubes without adapters. Up to fifteen racks of ten samples can be loaded into the input cue for a total of 150 samples.     Manual Open Tube Sampler (MOTS) The manual open tube sampler is located on the front of the analyzer. To aspirate a sample using the manual open tube sampler (MOTS), the sample probe is first immersed into the specimen.  The sampling process is initiated when the aspirate plate located directly behind the sample probe is pressed.    Manual Closed Tube Sampler (MCTS) To aspirate a sample using the manual closed tube sampler (MCTS), the capped vial is inserted upside down into the centering collar.  The sample tube is then pressed down into the centering collar.  The downward pressure on the centering collar causes the needle to push up, puncture the tube closure and aspirate the sample. On the first status line, the sample ID, sample type and test requested are displayed on the left side of the screen. On the right side of status line one, icons can be present indicating that service is due or the status of an attached Autoslide.  The left side of the second status line, indicates the analyzer status and the right side can display system messages and icons indicating the severity of the message.  Each of the Menu buttons on the top of the screen displays a menu of system functions. The menu displays when the pointer is moved over the button.  When selecting an item on a menu, the corresponding window opens, the button displays as pressed and the other functions on the menu display as tabs arranged beneath the status lines.  Tabs correspond to items on the active Menu button. You  can switch to any of the functions by selecting the corresponding tab. When an option from a different menu is selected, the tabs change to reflect those of the active menu.   Left-side buttons open windows within a Tab.  Some tabs contain more than one window.  The Help button provides a link to the Operator's Guide and additional information about the current screen. .. Shortcut Keys provide quick links to frequently used menu items. Up to six short cut keys can be customized to fit the operator's needs.   Bottom buttons activate commands or manipulate the contents of the window.
