Calibration includes reviewing calibration status, processing a calibration, and reviewing calibration data. This clinical laboratory training qualifies for continuing education units.

To manually create a calibration order: At the Command Bar, select Calibration Select the Calibration Order tab Select a test Select a reagent lot Select a calibrator lot Select Save The system can be configured to automatically create a calibration order when calibration samples are loaded on the system. To configure automatic calibration ordering: Select Setup from the Command Bar Select the System Configuration tab Select System Settings Enable the Automatic Calibration Order Settings Review the calibration data The ADVIA Centaur XPT system uses a two-point calibration system: Uses low and high calibrators Eliminates the need to measure a full standard curve or calibrate every time the assay is performed Screens used to review calibration data: Calibration Results Overview: "Valid" indicates that the Calibration Evaluation Criteria have passed Calibration Summary: "Current Calibration" indicates that the Calibration Evaluation Criteria have passed Discuss the accept option Accepting a Calibration: Required when the calibration status is "Pending Accept" status on the Calibration Results Overview screen "Pending Accept" calibrations are outside of observed ranges but within defined ranges Observed ranges: After 4 calibrations of the same reagent lot and calibrator lot combination, the system calculates observed ranges Defined ranges: The values for Slope, Ratio, Low Cal Deviation and High Cal Deviation entered from the Master Curve card To accept, select the Accept button for that calibration result on the Calibration Data screen After accepting, the calibration status changes to "Valid Operator Accepted" Select Next to continue. Tab TitleTextCalibration Summary Access the Calibration Summary screen by selecting Calibration from the Command Bar, and then selecting the Calibration Summary tab. The Calibration Summary screen displays the Calibration Status of tests. The filter button can be used to limit the display to only tests for reagents that are onboard, or to selected tests.      Reagent Status Access the Reagent Status screen by selecting Reagents from the Command Bar, and then selecting the Status tab. The Reagent Status screen displays information for all of the reagents onboard the system, including the calibration status and calibration expiration date. Pass the card under the scanner until the blue light points at the center of the 2D barcode. The scanner beeps once and the green light on the scanner flashes once after successfully reading the first 2D barcode. Note: You can also scan the 2D barcode by removing the scanner, holding the MC DEF card and pointing the blue light at the center of the 2D barcode. Scan the Second 2D BarcodeHTML Scan the second 2D barcode. The scanner beeps twice and the green light on the scanner flashes twice after successfully reading the second 2D barcode.     Scan the Remaining 2D Barcodes (if applicable)HTML   Scan the third and fourth 2D barcodes if included on the MCDEF card. The scanner beeps 3 times and 4 times after successfully reading the third and fourth 2D barcodes.     Select SaveHTML Select Save in the confirmation window to save the master curve and test definition and close the window. The master curve will now be listed on the Master Curve Definition screen. Base ImageHotspotsText BlocksImage File The Low and High Cal Deviations are a measure of how well the high and the low calibrator RLUs fit the Master Curve.  Slope is a comparison of high and low calibrator RLUs to the Master Curve. Ratio is a comparison of low and high calibrator RLUs. Calculated CV is the precision of the calibrator replicates based on RLUs. Defined ranges are the values for Slope, Ratio, Low Cal Deviation and High Cal Deviation derived when the MC DEF card is scanned for a specific reagent lot. Since these ranges are reagent lot specific, the relatively narrow limits of the ranges ensure quality calibration monitoring.   Acceptable CV is the maximum CV that is acceptable for a valid calibration. After performing 4 calibrations for a reagent lot and calibrator lot combination, the system calculates Observed Ranges for Slope, Ratio, Low Cal Deviation and High Cal Deviation. When you perform subsequent calibrations using the same reagent lot and calibrator lot combination, the system compares results to the defined ranges from the Master Curve definition and also the observed ranges determined by the system. The system recalculates observed ranges with each valid calibration. Critical Precision determines whether an outlier is too far out to automatically exclude it.
